生物實(shí)驗(yàn)稀釋過(guò)程分步進(jìn)行(例如 1ul母液,可以先加入1ul生理鹽水或者培養(yǎng)基,混合均勻沒(méi)有析出之后再加入2ul稀釋液,依次類推,逐漸稀釋。如果有析出及時(shí)超聲復(fù)溶。)在稀釋時(shí)要確保工作液中 DMSO 的終濃度盡量在0.1%以下,最高不要超過(guò)0.5%,并設(shè)置相應(yīng)濃度的DMSO對(duì)照組。DMSO熔點(diǎn)是18.5℃,一旦稀釋用的培養(yǎng)基溫度較低,DMSO結(jié)晶,藥物就會(huì)析出。所以稀釋過(guò)程注意一下溫度控制。
